B8S8 CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy developed by Outpace Bio for the treatment of mesothelin-positive solid tumors and hematologic malignancies. It utilizes a fully-human single-domain binder (VHH) targeting the membrane-distal Region 1 (RI) of the mesothelin (MSLN) protein. The construct features an optimized spacer (B8S8) designed using OutSpacer™ technology to achieve an ideal immune synapse geometry (~200 Å), which enhances potency and resistance to inhibition by soluble MSLN or MUC16. Additionally, the CAR is expressed under the control of a novel stim-on promoter (OP1) that dynamically regulates CAR expression in response to antigen stimulation, aiming to reduce T-cell exhaustion and improve durability compared to constitutive expression.
