BA-5 is a novel barbituric acid derivative investigated for the treatment of hepatocellular carcinoma (HCC) and sorafenib-resistant HCC. It exerts its antitumor effects by significantly inhibiting cell viability in a dose- and time-dependent manner. Its mechanism of action involves decreasing the phosphorylation of AKT and p70s6k, activating apoptosis through increased cleavage of PARP and caspase-7, and inhibiting cell migration by reducing Vimentin protein expression. BA-5 has shown synergistic effects when combined with low doses of sorafenib or regorafenib in HCC and sorafenib-resistant HCC cells, respectively.
