Drug intelligence / Profile preview
BA1203 is an investigational, next-generation, masked PD-1/IL-2 antibody-cytokine fusion protein (prodrug) developed by Boan Biotech for the treatment of advanced or metastatic solid tumors. It is designed to simultaneously achieve immune checkpoint blockade and localized immune activation within the tumor microenvironment (TME). BA1203 features a high-affinity, bivalent PD-1-targeting antibody fused to a masked IL-2 cytokine. The IL-2 component is designed as a prodrug that is selectively activated within the TME, thereby minimizing peripheral IL-2 activity and reducing systemic toxicity while maintaining high activity at the IL-2α receptor. Preclinical studies have demonstrated superior antitumor activity compared to existing PD-1/PD-L1 inhibitors, particularly in models with low or no response to standard checkpoint blockade. A Phase I clinical trial is evaluating its safety, tolerability, pharmacokinetics, and preliminary efficacy in patients with advanced solid tumors.
