BA1302 is a novel, investigational antibody-drug conjugate (ADC) targeting CD228, developed by Shandong Boan Biotechnology. It is the first and only CD228-directed ADC in clinical development worldwide. The drug consists of a human monoclonal antibody that specifically binds to the membrane-bound form of CD228 (melanotransferrin), sparing the soluble form and thus minimizing off-target toxicity. The antibody is conjugated via interchain cysteine bonds to a cytotoxic payload—monomethyl auristatin E (MMAE)—using a cleavable hydrophilic linker designed for high stability both in vitro and in vivo[1][5][2]. This design enables potent internalization into tumor cells, resulting in cell death through microtubule disruption, with demonstrated bystander effect and robust antitumor activity across multiple solid tumor models[1][5][4]. Preclinical studies have shown favorable pharmacokinetics, prolonged half-life, good safety/tolerability profiles in animal models, and significant efficacy against tumors expressing varying levels of CD228[2][5]. BA1302 has received orphan drug designation from the US FDA for squamous non-small cell lung cancer (NSCLC) and pancreatic cancer. It is currently being evaluated in phase 1 clinical trials for advanced solid tumors including NSCLC (squamous), pancreatic adenocarcinoma, breast cancer, melanoma, adenocarcinoma, among others[1][7][8].
