A combination therapy of **BA3071 (evalstotug)**, a conditionally active biologic (CAB) **anti–CTLA-4 monoclonal antibody**, and **nivolumab**, a programmed cell death protein 1 (**PD-1**) inhibitor. BA3071 is designed to selectively block CTLA-4 in the acidic tumor microenvironment, reducing off-tumor and systemic immune-related adverse effects while retaining antitumor efficacy similar to other anti–CTLA-4 agents. When used in combination with nivolumab, the regimen aims to enhance immune-mediated tumor control by dual checkpoint inhibition. The combination is being evaluated for advanced solid tumors, including but not limited to melanoma and non-small cell lung cancer (NSCLC) with various mutations, in a multi-phase clinical program. The approach specifically targets patients naïve to anti–CTLA-4 therapy, most of whom have already failed prior PD-1 inhibitor therapy[1][4][5].
