Overview

**BA3142** is an IND-ready, preclinical dual conditionally active biologic T-cell engager developed by BioAtla for solid tumors. It is a tetravalent bispecific antibody that conditionally binds the tumor-associated antigen B7-H3 and CD3 on T cells under acidic conditions characteristic of the tumor microenvironment, while exhibiting lower binding activity under normal physiological conditions. By co-localizing CD3-positive T cells with B7-H3-expressing tumor cells, BA3142 is designed to induce tumor-directed T-cell cytotoxicity while widening the therapeutic window relative to constitutively active T-cell engagers. Preclinical antitumor activity has been reported in humanized mouse models of melanoma and pharyngeal cancer.
