Overview

BA3181 is a novel conditionally active biologic (CAB) antibody-drug conjugate (ADC) designed to target the Epithelial Cell Adhesion Molecule (EpCAM). Developed by BioAtla, BA3181 utilizes proprietary CAB technology, which allows the antibody to bind to its target preferentially under the unique conditions of the tumor microenvironment (such as low pH) while remaining inactive in normal physiological conditions. This conditional activation is intended to improve the safety profile and therapeutic index of EpCAM-targeted therapy, which has historically been limited by off-target toxicity in healthy epithelial tissues. EpCAM is highly expressed in a variety of solid tumors and cancer stem cells, making it an attractive target for ADC-mediated delivery of cytotoxic agents. Although BA3181 entered Phase 1 clinical trials for advanced solid tumors, the program was subsequently terminated by the developer to prioritize other assets in their pipeline.
