Overview

BA3182 is a conditionally active biologic (CAB) bispecific T-cell engager antibody developed by BioAtla for the treatment of advanced adenocarcinoma and other solid tumors. The drug is designed to target both the epithelial cell adhesion molecule (EpCAM), which is commonly overexpressed in many epithelial cancers, and CD3ε on T cells. Its mechanism leverages dual binding sites for EpCAM and CD3ε that are engineered to bind specifically under the acidic conditions of the tumor microenvironment (TME), with reduced binding at normal physiological pH. This conditional activity aims to enhance selectivity for tumor cells while minimizing off-tumor toxicity. BA3182 is being investigated in clinical trials for multiple cancer types including adenocarcinoma, non-small cell lung cancer, small-cell lung cancer, colorectal cancer, prostate cancer, colon cancer, pancreatic cancer, triple-negative breast cancer, and was also previously under development for ovarian cancer[1][2][6].
