BACE2 siRNA is a small interfering RNA (siRNA) designed to silence the expression of the beta-site amyloid precursor protein-cleaving enzyme 2 (BACE2). BACE2 is a protease that, under physiological conditions, typically acts as an alpha-secretase, cleaving the amyloid precursor protein (APP) within the amyloid-beta (Aβ) sequence to prevent the formation of toxic Aβ plaques. In research contexts, such as studies involving human brain microvascular endothelial cells (BMECs), BACE2 siRNA is utilized to investigate the enzyme's role in APP metabolism and its potential involvement in Alzheimer's disease pathogenesis. Experimental results indicate that knockdown of BACE2 reduces the secretion of the neurotrophic product soluble APP alpha (sAPPα) and may influence the expression of other proteins like BACE1 and ADAM10, suggesting that BACE2 inhibition could impair protective pathways in the brain.
