Bacillus subtilis PXN21 is a specific probiotic bacterial strain being developed by ADM Protexin for the treatment of Parkinson's disease. Originally isolated from the commercial probiotic Bio-Kult, this strain consists of live bacterial spores that have demonstrated the ability to inhibit, delay, and reverse the aggregation of alpha-synuclein, a hallmark protein in Parkinson's pathology. Preclinical studies in Caenorhabditis elegans suggest the mechanism involves the activation of the DAF-16/FOXO protective pathway and alterations in host sphingolipid metabolism, potentially mediated by bacterial metabolites and biofilm formation in the gut. It is currently being evaluated in Phase 2 clinical trials to assess its impact on the gut microbiome and blood biomarkers in Parkinson's patients.
