Overview

Baclofen is a small molecule skeletal muscle relaxant and antispasmodic agent, primarily used to treat spasticity associated with multiple sclerosis, spinal cord injury, or other spinal cord diseases. It acts as a specific agonist at the gamma-Aminobutyric acid type B receptor (GABA-B receptor), reducing excitatory neurotransmission in the central nervous system. This leads to decreased muscle spasms, cramping, and tightness. While originally developed for epilepsy, its main clinical use is now for severe spasticity of cerebral or spinal origin in both adults and children over 12 years old. It may also be used off-label for alcohol use disorder and opioid withdrawal symptoms[1][2][3][4][5][6].
