bacTRL-IL-12 is a novel, genetically engineered bacterial therapy designed to deliver interleukin-12 (IL-12) directly to tumor sites. The therapy uses a modified strain of Bifidobacterium longum that encodes the IL-12 transgene. Upon administration, these bacteria selectively colonize hypoxic regions within solid tumors and locally release IL-12, aiming to stimulate an anti-tumor immune response while minimizing systemic toxicity[1][3]. IL-12 is a cytokine known for its ability to activate both innate and adaptive immunity by promoting T helper 1 (Th1) responses, enhancing cytotoxic T lymphocyte and natural killer cell activity, and increasing interferon-gamma production[5][6]. The primary indication for bacTRL-IL-12 has been advanced solid tumors. Development was initially led by IQVIA Ltd., but the global highest R&D status is currently listed as discontinued[2].
