BAFF CAR T (LUM-001) is an investigational, trispecific chimeric antigen receptor (CAR) T-cell therapy developed by researchers at UH Seidman Cancer Center and Case Western Reserve University in partnership with Luminary Therapeutics. Unlike traditional CAR-T therapies that target a single antigen, this product utilizes the B-cell activating factor (BAFF) ligand to simultaneously target three distinct receptors: B-cell activating factor receptor (BAFF-R), B-cell maturation antigen (BCMA), and transmembrane activator and CAML interactor (TACI). This multi-target approach is specifically designed to overcome antigen escape, a common mechanism of resistance where cancer cells downregulate or stop expressing a single targeted protein. The therapy is currently being evaluated in a Phase 1 clinical trial for patients with relapsed or refractory multiple myeloma who have received three or more prior lines of therapy. The manufacturing process utilizes electroporation for gene delivery and is conducted at the Wesley Center for Immunotherapy.
