Drug intelligence / Profile preview
BAFF CAR T-cells (B-cell-activating factor chimeric antigen receptor T-cells) are genetically engineered T-cells designed to express a chimeric antigen receptor utilizing the BAFF ligand (or a truncated trimeric BAFF ligand) as the antigen-binding domain. Unlike traditional single-target CAR T-cells (such as anti-CD19 or anti-BCMA), BAFF CAR T-cells target three distinct cognate receptors of the BAFF ligand: BAFF receptor (BAFF-R/TNFRSF13C), transmembrane activator and CAML interactor (TACI/TNFRSF13B), and B-cell maturation antigen (BCMA/TNFRSF17). This multi-antigen targeting strategy is designed to prevent antigen escape, a common mechanism of resistance and relapse in B-cell malignancies. BAFF CAR T-cells are being developed for the treatment of B-cell malignancies, including non-Hodgkin lymphoma (NHL), chronic lymphocytic leukemia (CLL), and multiple myeloma (MM), as well as autoimmune diseases such as systemic lupus erythematosus (SLE) and membranous nephropathy (MN).
