BAFFR-CAR-NK cell therapy is an investigational allogeneic (off-the-shelf) cell therapy being developed by PeproMene Bio. The therapy utilizes natural killer (NK) cells engineered with a chimeric antigen receptor (CAR) that specifically targets the B-cell activating factor receptor (BAFFR). BAFFR is a member of the tumor necrosis factor receptor superfamily and is essential for the survival and maturation of B cells, with expression limited almost exclusively to the B-cell lineage. By targeting BAFFR, this therapy aims to eliminate malignant or pathogenic B cells while potentially offering a safer profile and broader accessibility compared to autologous CAR-T therapies, as NK cells do not require HLA matching and carry a lower risk of graft-versus-host disease. It is currently in preclinical development for the treatment of B-cell malignancies and autoimmune disorders.
