BAH421 is a dual-targeting chimeric antigen receptor (CAR) T-cell therapy developed by Essen Biotech for the treatment of B-cell acute lymphoblastic leukemia (B-ALL). The therapy utilizes autologous T cells engineered to express CARs that simultaneously recognize CD19 and CD22, two antigens highly expressed on the surface of malignant B cells. This bispecific approach is designed to mitigate the risk of tumor escape through antigen loss, a common resistance mechanism observed with single-target CD19 CAR-T therapies. Upon binding to either CD19 or CD22, the engineered T cells are activated to induce direct cytotoxic killing of the leukemic cells. BAH421 is currently being evaluated in Phase 2 clinical trials.
