Bakuchicin is a natural furanocoumarin compound with anti-inflammatory properties, primarily investigated for its potential in treating allergic airway inflammation and asthma. It exerts its effects by regulating macrophage activation and polarization. Specifically, it inhibits the production of pro-inflammatory cytokines and nitric oxide by suppressing the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). It also modulates signaling pathways including Akt, MAPKs, and NF-κB. Furthermore, bakuchicin suppresses IL-4-induced M2 macrophage polarization by inhibiting markers such as arginase-1, YM-1, and Fizz-1, and by regulating sirtuin 2 levels.
