Overview

Balomenib is an oral, reversible small-molecule inhibitor of the menin (MEN1) protein-protein interaction (PPI), currently under development by Clywedog Therapeutics. The drug is designed to disrupt pathological menin-dependent transcriptional programs that regulate pancreatic beta-cell function and survival. By targeting these pathways, balomenib aims to preserve and restore beta-cell mass and capacity, offering a potential disease-modifying approach for the treatment of type 2 diabetes (T2D) and type 1 diabetes (T1D). As of late 2025, it has entered Phase 1b clinical testing for type 2 diabetes.
