Overview

Balstilimab + zalifrelimab is a combination of two investigational monoclonal antibodies developed for cancer immunotherapy, particularly in advanced or recurrent/metastatic cervical cancer. Balstilimab is a fully human IgG4 monoclonal antibody that targets and blocks programmed death protein 1 (PD-1), preventing its interaction with PD-L1 and PD-L2, thereby enhancing T-cell mediated immune responses against tumor cells. Zalifrelimab is a fully human IgG1 monoclonal antibody that targets cytotoxic T-lymphocyte–associated antigen 4 (CTLA-4), blocking its interaction with CD80 and CD86 to further promote immune activation. The dual checkpoint blockade aims to overcome tumor-induced immunosuppression by simultaneously inhibiting two key negative regulators of immune activation. This combination has shown promising clinical activity and manageable safety in phase II trials for patients with recurrent or metastatic cervical cancer who have relapsed after platinum-based therapy[2][3][5][7].
