Baminercept is a recombinant fusion protein that acts as a soluble lymphotoxin beta receptor (LTβR) antagonist. It is designed to inhibit the lymphotoxin pathway by binding and neutralizing ligands for the LTβR, thereby blocking downstream signaling involved in inflammation and lymphoid tissue organization. Baminercept was developed primarily for autoimmune diseases such as primary Sjögren's syndrome and rheumatoid arthritis. Despite evidence of biological activity—such as reduced plasma CXCL13 levels and changes in B/T cell populations—clinical trials did not demonstrate significant efficacy over placebo in improving disease symptoms. The drug has also been investigated for ulcerative colitis, Crohn's disease, and multiple sclerosis but development has been discontinued for these indications[1][3][5][6].
