Overview
Baminercept alfa + methotrexate is a combination therapy that was investigated for the treatment of moderate to severe rheumatoid arthritis (RA), particularly in patients with inadequate response to standard therapies. Baminercept alfa is a fusion protein (LTβR-Ig) targeting the lymphotoxin-β and LIGHT pathways, crucial components of the TNF superfamily implicated in autoimmune inflammation. Methotrexate is a well-established disease-modifying antirheumatic drug (DMARD) that inhibits lymphocyte proliferation via folate antagonism. Together, this combination aims to modulate both lymphocytic and stromal contributions to chronic joint inflammation. Baminercept alfa was developed by Biogen Idec and acts by blocking the formation and maintenance of aberrant lymphoid structures involved in chronic autoimmune responses, whereas methotrexate broadly inhibits immune cell proliferation[1][2][5].
