Bamlanivimab is a fully human IgG1κ monoclonal antibody that targets the spike (S) protein of SARS-CoV-2, the virus responsible for COVID-19. It was developed through high-throughput screening of B-cells from a convalescent COVID-19 patient. Bamlanivimab binds to the receptor-binding domain (RBD) of the spike protein at a site overlapping with the ACE2 binding site, thereby blocking viral attachment and entry into human cells. This neutralizing action is intended to reduce viral load and prevent progression to severe disease in patients with mild-to-moderate COVID-19 who are at high risk for complications. The drug was developed by AbCellera Biologics and Eli Lilly and received emergency use authorization (EUA) from the US FDA in November 2020; however, this authorization has since been revoked due to reduced efficacy against emerging variants[1][7][8].
