Bapotulimab is a fully human IgG2-kappa monoclonal antibody that targets immunoglobulin-like domain containing receptor 2 (ILDR2), a novel immune checkpoint protein. By binding to and inhibiting ILDR2, bapotulimab blocks its immunosuppressive activity, thereby preventing ILDR2-mediated inhibition of T-cell activities. This leads to the stimulation of cytotoxic T lymphocyte (CTL) responses against tumor cells and enhances anti-tumor immunity. Bapotulimab has demonstrated anti-tumor activity as monotherapy in preclinical models and shows synergistic effects when combined with other cancer therapies. It is being developed primarily for the treatment of solid tumors, including head and neck squamous cell carcinoma, with ongoing phase I clinical trials evaluating its safety and efficacy both as monotherapy and in combination with pembrolizumab[1][2][3][4][7].
