Overview

BAQ13-NP is a novel nanoparticle-encapsulated bisaminoquinoline derivative designed for the treatment of glioblastoma (GBM). Developed by researchers at UC Davis, it aims to overcome the limitations of traditional aminoquinolines, such as hydroxychloroquine, by enhancing potency and blood-brain barrier (BBB) penetration. The mechanism of action involves the inhibition of autophagy, evidenced by the accumulation of LC3B and p62, and the suppression of IL-6-dependent STAT3 phosphorylation in tumor cells and the immune microenvironment. Preclinical studies in orthotopic murine glioma models (GL261 and CT2A) demonstrated significant survival improvements and reduced tumor growth following systemic administration. BAQ13-NP has received Investigational New Drug (IND) approval and is transitioning into Phase I clinical trials.
