Overview

BAT7104 is a bispecific monoclonal antibody developed by Bio-Thera Solutions that targets and inhibits both the PD-1/PD-L1 and CD47/SIRP-α pathways. It is designed with an "imbalanced" binding affinity—finely tuned for CD47 and high for PD-L1—to selectively block CD47 on tumor cells that co-express PD-L1, thereby minimizing red blood cell depletion. Mechanistically, it blocks immune checkpoint signaling (PD-1/PD-L1) to enhance T-cell activation while also inhibiting the "don't eat me" signal (CD47/SIRP-α), promoting macrophage-mediated phagocytosis of tumor cells. Preclinical studies have shown synergistic anti-tumor activity and favorable safety in non-human primates. BAT7104 is currently being evaluated in Phase 1 clinical trials for advanced solid tumors and lymphoma[1][2][3][4][5][7].
