BAT8010 is an antibody-drug conjugate (ADC) developed by Bio-Thera Solutions for the treatment of solid tumors. It consists of a monoclonal antibody targeting human epidermal growth factor receptor 2 (HER2), which is overexpressed in various cancers, linked to the camptothecin analog exatecan—a potent topoisomerase I inhibitor. The ADC uses a cleavable but systemically stable linker and has a high drug-to-antibody ratio (DAR). Upon binding to HER2-expressing tumor cells, BAT8010 is internalized and releases its cytotoxic payload, leading to cell death; the released payload can also kill neighboring cancer cells via a bystander effect. This mechanism aims to overcome tumor heterogeneity and improve efficacy in advanced or metastatic solid tumors such as breast cancer, gastric cancer, biliary tract neoplasms, colorectal cancer, non-small cell lung cancer, and others. As of June 2024, BAT8010 is in Phase 1/2 clinical trials for these indications[1][2][3][6][7].
