BAT8013 is an investigational antibody-drug conjugate (ADC) developed by Bio-Thera Solutions, targeting Protein Tyrosine Kinase 7 (PTK7). PTK7 is a cell surface protein that is frequently overexpressed in various solid malignancies, including non-small cell lung cancer (NSCLC), esophageal cancer, and breast cancer, while maintaining limited expression in normal tissues. The ADC is composed of a humanized anti-PTK7 monoclonal antibody conjugated to a proprietary topoisomerase I inhibitor payload via a cleavable linker. The mechanism of action involves the selective binding of the antibody to PTK7 on the surface of tumor cells, followed by internalization of the complex. Once inside the cell, the cytotoxic payload is released, where it inhibits DNA topoisomerase I, leading to double-strand DNA breaks and subsequent apoptosis. BAT8013 is currently being evaluated in Phase I/II clinical trials in China for the treatment of patients with advanced solid tumors who have failed or lack standard therapies.
