Overview

Bavituximab is an investigational monoclonal antibody targeting phosphatidylserine (PS), a phospholipid exposed on tumor blood vessels and tumor cells that creates an immunosuppressive environment. By binding PS, bavituximab reverses this immunosuppression and promotes immune activation against tumors. Ipilimumab is a monoclonal antibody that targets CTLA-4, a checkpoint receptor on T cells, thereby activating the immune system to attack cancer cells. The combination of bavituximab and ipilimumab aims to enhance anti-tumor immune responses by combining upstream checkpoint inhibition (bavituximab) with downstream checkpoint blockade (ipilimumab). This combination has been investigated in clinical trials for advanced melanoma, showing potential for improved efficacy compared to ipilimumab alone. The combination trial was conducted as an open-label Phase Ib study at the University of Texas Southwestern Medical Center[1][2][4].
