Bavunalimab (also known as XmAb841, XmAb-22841, or pavunalimab) is a bispecific monoclonal antibody designed to simultaneously target and inhibit two immune checkpoint receptors: CTLA-4 (Cytotoxic T-Lymphocyte Associated Protein 4) and LAG-3 (Lymphocyte Activation Gene 3). This antibody was developed by Xencor for use as an immunotherapy in oncology, specifically as an antineoplastic agent for advanced solid tumors and malignant melanoma. The bispecific molecule is engineered for antibody-dependent cell cytotoxicity and functions by inhibiting both CTLA-4 and LAG-3 pathways, aiming to enhance anti-tumor immune responses. Bavunalimab advanced to Phase 1 clinical studies in several cancer indications but was discontinued following underwhelming early-phase trial data[1][5][7][8][11].
