BAY 2287411 is a targeted thorium conjugate (TTC) consisting of the alpha-emitting radioisotope thorium-227 (227Th) conjugated to the anti-mesothelin monoclonal antibody anetumab via a 3,2-HOPO chelator. Developed by Bayer in collaboration with Lumiphore and ImmunoGen, the drug is designed to deliver high-energy alpha radiation specifically to tumor cells expressing mesothelin, a cell-surface glycoprotein overexpressed in several cancers including mesothelioma and ovarian carcinoma. Alpha particles induce complex double-strand DNA breaks that are difficult for cells to repair, leading to cell death. BAY 2287411 has been investigated in Phase 1 clinical trials for the treatment of advanced mesothelin-positive malignancies.
