Overview

BAY 2315497 (Bayer PSMA Targeted 227Th-Conjugate) is a targeted thorium conjugate (TTC) developed by Bayer in collaboration with Lumiphore and Progenics (now Lantheus). It consists of a humanized anti-prostate-specific membrane antigen (PSMA) monoclonal antibody conjugated to a 3,2-HOPO chelator that complexed with the alpha-emitting radioisotope thorium-227. The drug is designed to treat prostate cancer by binding to PSMA-expressing tumor cells and delivering high-energy alpha particles directly to the tumor site. These alpha particles cause lethal double-stranded DNA breaks, leading to cell death while minimizing damage to surrounding healthy tissue due to their short range. BAY 2315497 is currently in early clinical development for the treatment of metastatic castration-resistant prostate cancer.
