BAY 2501293 is a molecular glue, part of a class of compounds termed velcrins, developed by Bayer. It acts by promoting the formation of a complex between phosphodiesterase 3A (PDE3A) and the ribonuclease Schlafen 12 (SLFN12). This molecular glue mechanism activates SLFN12, which then specifically cleaves the leucine tRNA (tRNALeu(TAA)), resulting in the inhibition of protein translation and the induction of apoptosis. BAY 2501293 has shown efficacy in preclinical glioblastoma models, particularly those with high expression of the biomarker pair PDE3A and SLFN12.
