BAY 3498264 + sotorasib is an investigational combination therapy composed of BAY 3498264, a small molecule inhibitor of **son of sevenless 1 (SOS1)**, and **sotorasib**, an already-approved targeted inhibitor of mutant **KRAS G12C**. The combination aims to improve clinical outcomes for patients with advanced solid tumors harboring **KRAS G12C mutations** by inhibiting two related drivers in the RAS pathway: sotorasib directly inhibits mutant KRAS G12C, while BAY 3498264 blocks the function of SOS1, a protein upstream in the RAS signaling cascade that promotes KRAS activation. This dual inhibition is proposed to produce deeper or more durable responses than sotorasib alone, as resistance and cancer progression remain challenges with sotorasib monotherapy. The combination is currently being tested in a first-in-human clinical trial for safety, maximum tolerated dose, and initial efficacy in adults with advanced KRAS G12C-mutated cancers[1][3][5].
