Overview

BAY 79-4620 is an investigational antibody-drug conjugate (ADC) developed for the treatment of solid tumors that overexpress carbonic anhydrase IX (CAIX). The drug consists of a monoclonal antibody (3ee9) targeting human CAIX linked to the cytotoxic agent monomethyl auristatin E (MMAE) via a self-immolative enzyme-cleavable linker. CAIX is a cell surface glycoprotein highly expressed in various tumor types and associated with hypoxia and poor prognosis. Upon binding to CAIX-expressing tumor cells, the ADC is internalized and releases MMAE intracellularly to inhibit tubulin polymerization, leading to mitotic arrest and apoptosis. Preclinical studies demonstrated potent antitumor activity in xenograft models with partial or complete tumor regression after single doses. Clinical development reached phase I trials in patients with advanced solid tumors but was discontinued[1][2][3][4][5][6].
