BAY-87-2243 is a highly potent and selective small molecule inhibitor of hypoxia-inducible factor 1 (HIF-1). It inhibits HIF-1 reporter gene activity and CA9 protein expression at nanomolar concentrations. The drug acts by blocking mitochondrial complex I activity, thereby reducing HIF protein levels under hypoxic conditions. This mechanism leads to suppression of HIF target gene expression and inhibition of HIF-1α protein accumulation in cancer cells. In preclinical models, BAY-87-2243 demonstrated dose-dependent antitumor efficacy without significant toxicity or weight loss. It was developed for the treatment of malignancies but clinical development was terminated due to drug-related adverse events[1][5][7][8].
