Overview

BAY2253651 is a nasally applied small molecule drug developed as a genioglossus muscle activator for the treatment of obstructive sleep apnoea (OSA). Its mechanism of action involves selective antagonism of TASK-1 and TASK-3 potassium channels in the pharyngeal mucosa, which is intended to stimulate upper airway dilator muscle activity via pharyngeal mucosal receptor stimulation. The goal was to reduce upper airway collapsibility during sleep and thereby improve OSA symptoms. Clinical trials demonstrated that while BAY2253651 was safe and well-tolerated, it did not show significant therapeutic efficacy in reducing OSA severity compared to placebo[1][4][5][7]. The drug's development for this indication has been discontinued.
