Overview

BAY2416964 is a potent, orally bioavailable small molecule antagonist of the aryl hydrocarbon receptor (AhR). It specifically binds to and inhibits AhR activation, thereby preventing AhR-mediated signaling. The drug is designed to counteract the immunosuppressive effects mediated by AhR ligands in the tumor microenvironment, restoring immune cell function and enhancing proinflammatory activity of antigen-presenting cells and T cells while reducing immunosuppressive myeloid cell activity. BAY2416964 is being developed primarily for the treatment of advanced solid tumors, including non-small-cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), colorectal cancer (CRC), and other solid tumors. Preclinical studies have shown that it can restore immune responses against tumor cells and enhance cytotoxic T cell activity[1][2][3][7].
