Overview

BAY3401016 is a small molecule soluble guanylate cyclase (sGC) activator developed by Bayer AG, currently in early-phase clinical development for the treatment of Alport syndrome. Alport syndrome is a genetic condition characterized by progressive kidney disease, hearing loss, and eye abnormalities, often driven by mutations in Type IV collagen genes that lead to glomerular basement membrane dysfunction and subsequent fibrosis. BAY3401016 aims to address the underlying pathophysiology by activating sGC, an enzyme that produces cyclic guanosine monophosphate (cGMP). Unlike sGC stimulators, which require the presence of nitric oxide and a reduced heme group, sGC activators can stimulate the enzyme even when it is oxidized or heme-deficient—conditions often found in states of high oxidative stress. By restoring cGMP signaling, the drug is intended to exert anti-fibrotic and anti-inflammatory effects, potentially slowing the progression of renal damage in Alport syndrome patients.
