Overview

BAY86-4980 is an antibody-drug conjugate (ADC) developed by Bayer for the treatment of solid tumors. It consists of a human monoclonal antibody targeting C4.4a (also known as Ly6/PLAUR domain-containing protein 3 or LYPD3), a cell surface protein highly expressed in squamous cell carcinomas, conjugated to the cytotoxic maytansinoid payload DM4 via a disulfide-containing SPDB linker. The drug was designed to deliver the potent microtubule inhibitor directly to tumor cells expressing C4.4a, leading to cell cycle arrest and apoptosis. Clinical development reached Phase 1 for indications including non-small cell lung cancer and head and neck squamous cell carcinoma, but the program was discontinued.
