Bazedoxifene is a third-generation selective estrogen receptor modulator (SERM) developed by Pfizer. It acts as an estrogen receptor agonist or antagonist depending on the tissue type, functioning primarily as an antagonist in uterine and breast tissues and as an agonist in bone. Bazedoxifene decreases bone resorption, reduces biochemical markers of bone turnover to premenopausal levels, and increases bone mineral density (BMD), thereby reducing fracture risk. It is used alone or in combination with conjugated estrogens for the prevention of postmenopausal osteoporosis and for the treatment of moderate to severe vasomotor symptoms associated with menopause[2][3][5][6]. In combination therapy (as Duavee/Duavive), it also helps reduce the risk of endometrial hyperplasia that can be caused by estrogens[4].
