Drug intelligence / Profile preview
BB-0001 is an optimized, alkaloid-derived small molecule entry inhibitor being developed by the Translational Health Science and Technology Institute (THSTI) as a broad-spectrum antiviral candidate for the treatment of Dengue virus (DENV) infections [E0046]. BB-0001 exhibits potent antiviral activity against all four DENV serotypes (DENV1–4) by blocking viral entry, inhibiting RNA-dependent RNA polymerase (RdRp)-mediated replication, and enhancing host antiviral signaling pathways [E0046]. In addition to its direct antiviral effects, BB-0001 demonstrates immunomodulatory properties by suppressing the production of pro-inflammatory cytokines, such as IL-6 and TNF-α, in primary human macrophages [E0046]. Preclinical studies in mouse models have shown that BB-0001 possesses good oral bioavailability, a moderate half-life of approximately 6 hours, and the ability to significantly reduce viremia and prevent hepatic injury without observable toxicity [E0046].
