Overview

BB-025 is an investigational, rapid-acting reversal oligonucleotide being developed by Basking Biosciences as a **direct-acting reversal agent** for the RNA aptamer thrombolytic BB-031, which targets von Willebrand Factor for treatment of acute ischemic stroke and other acute thrombotic conditions.[1][7][8][9][12] BB-025 is designed as a complementary oligonucleotide that specifically binds to and neutralizes BB-031, forming a drug–drug complex that rapidly and durably reverses BB-031’s thrombolytic and antiplatelet effects within minutes after a single intravenous bolus, thereby providing an “on‑off switch” to control bleeding risk or enable urgent surgery.[1][3][6][7][8][9] A Phase 1 randomized, placebo-controlled, dose‑escalation study in healthy volunteers (NCT07202663) is evaluating the safety, pharmacokinetics, pharmacodynamics, and reversal performance of single-dose BB-025 alone and following BB-031, with BB-025 currently characterized as a coagulation factor–modulating antifibrinolytic oligonucleotide for blood coagulation disorders and as a supportive agent for BB-031–based thrombolytic therapy.[1][5][7][8][9][10][11]
