Overview

BB-1701 is an investigational antibody-drug conjugate (ADC) composed of a humanized anti-HER2 monoclonal antibody with the identical amino acid sequence as trastuzumab, conjugated to the cytotoxic payload eribulin via a cathepsin B-cleavable linker[3][6][7]. After binding to HER2 on tumor cells, BB-1701 is internalized and eribulin is released inside the cell, causing irreversible mitotic blockage and tumor cell death through inhibition of tubulin polymerization. The released eribulin can also exert bystander effects on neighboring tumor cells and modulate the tumor microenvironment[2][6][7]. BB-1701 has demonstrated promising antitumor activity in preclinical models and early-phase clinical trials for HER2-positive and HER2-low expressing solid tumors, including breast cancer and non-small cell lung cancer[4][5]. It is being developed by Bliss Biopharmaceutical in collaboration with Eisai.
