BB-5568 is a preclinical-stage small molecule inhibitor developed by BrodenBio (Broaden Bio) for the treatment of leukemia. It specifically targets the protein-protein interaction between Menin and KMT2A (Lysine Methyltransferase 2A, formerly known as MLL1). This interaction is a critical driver in the pathogenesis of specific leukemia subsets, particularly those characterized by KMT2A rearrangements (KMT2Ar) or NPM1 mutations. By disrupting the binding of Menin to the KMT2A complex, BB-5568 suppresses the transcription of pro-leukemogenic genes such as HOXA9 and MEIS1, leading to the differentiation of leukemic blasts and the inhibition of cell proliferation. The drug is currently being evaluated in preclinical models for acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL).
