BB1001 is a cyclic oligonucleotide/peptide decoy designed to inhibit the Signal Transducer and Activator of Transcription 3 (STAT3) protein. Developed by Bluedot Bio, BB1001 is formulated for intravenous (IV) administration. The drug acts by mimicking the DNA binding site of STAT3, thereby sequestering the transcription factor and preventing it from binding to genomic DNA and driving the expression of genes involved in tumor progression, inflammation, and fibrosis. BB1001 is currently in IND-enabling studies for various indications, including head and neck squamous cell carcinoma (HNSCC), non-small cell lung cancer (NSCLC), colorectal cancer (CRC), and rare STAT3 gain-of-function diseases such as Noonan syndrome. It is also being explored for inflammatory conditions like psoriasis and fibrosis.
