Overview

BB14 is a murine IgG1 monoclonal antibody directed against the **CD4** antigen found on the surface of helper T lymphocytes. Developed in the early 1990s, it was primarily investigated for the treatment of severe **psoriasis** and other T-cell mediated autoimmune disorders. The antibody works by binding to the CD4 receptor, which leads to the coating of circulating T cells, a reduction in the density of CD4 molecules on the cell surface (antigenic modulation/down-regulation), and a partial depletion of the CD4+ T-cell population. In clinical studies for psoriasis, BB14 administration resulted in significant clinical improvement, characterized by a reduction in the Psoriasis Area Severity Index (PASI) and decreased expression of inflammatory markers such as ICAM-1 and HLA-DR on keratinocytes.
