bb21217 is a second-generation autologous chimeric antigen receptor (CAR) T cell therapy targeting B-cell maturation antigen (BCMA), developed for the treatment of relapsed or refractory multiple myeloma. It uses the same CAR molecule as idecabtagene vicleucel (bb2121), but during ex vivo manufacturing, T cells are cultured with a PI3 kinase inhibitor (bb007) to enrich for memory-like phenotype T cells. This approach aims to enhance in vivo persistence and durability of response. The drug was originally developed by bluebird bio and Bristol Myers Squibb/Celgene; later development rights were acquired by Regeneron Pharmaceuticals. Clinical trials demonstrated promising efficacy and safety profiles in heavily pre-treated multiple myeloma patients, but development was discontinued after Phase 1 due to portfolio prioritization[1][4][5][7][8].
