Overview

BB5.1 is a rat-derived monoclonal antibody that specifically targets and inhibits murine complement component 5 (C5). By binding to the C5 alpha-chain with high affinity, it prevents the proteolytic cleavage of C5 into the pro-inflammatory anaphylatoxin C5a and the fragment C5b, which is required for the assembly of the membrane attack complex (MAC or C5b-9). Originally raised in C5-deficient mice, BB5.1 has been a cornerstone of complement research for over 30 years, serving as the primary tool for demonstrating the therapeutic potential of C5 blockade in mouse models of disease. This research provided the critical proof-of-concept that led to the development of humanized anti-C5 therapies, most notably eculizumab. BB5.1 is extensively utilized in preclinical studies to investigate complement-mediated pathology in conditions ranging from ischemia-reperfusion injury and organ transplant rejection to autoimmune and inflammatory disorders.
