Overview

BBI-10 is an oral, small-molecule, covalent inhibitor of the Stimulator of Interferon Genes (STING) pathway being developed for autoimmune, inflammatory, and rare genetic diseases in which aberrant STING activation drives type I interferon signaling. As a covalent STING inhibitor, BBI-10 is designed to bind irreversibly to STING and suppress downstream interferon and inflammatory cytokine production, with the goal of modulating pathogenic innate immune activation while preserving host defense. The program was initially developed by Carna Biosciences and subsequently advanced by Brickell Biotech (later Fresh Tracks Therapeutics), and remains in preclinical development for indications such as systemic lupus erythematosus, dermatomyositis, and nonalcoholic steatohepatitis.[1][4][11]
