Overview

BBI-355 is an orally available, potent, and selective small molecule inhibitor of checkpoint kinase 1 (CHK1), developed as the first ecDNA-directed therapy (ecDTx) for the treatment of oncogene-amplified cancers. CHK1 is a master regulator of DNA replication stress (RS), which frequently arises from oncogene amplification on extrachromosomal DNA (ecDNA) in cancer cells. By inhibiting CHK1, BBI-355 disrupts the repair of damaged DNA and exploits a synthetic lethal vulnerability in ecDNA-driven tumor cells, leading to preferential cancer cell death while sparing healthy cells. BBI-355 is currently being evaluated in Phase 1/2 clinical trials for patients with locally advanced or metastatic solid tumors harboring oncogene amplifications.
